An overview of the peptide synthesis

In organic chemistry, peptide synthesis is the process of peptides production, in which amino acids are linked through peptide bonds. This is the contrast of the biological process of producing long peptides. A number of peptides are significant commercial or pharmaceutical products that range from the dipeptide sugar substitute aspartame in order to clinically used hormones. This includes oxytocin, calcitonin, and adrenocorticotropin.

Protein synthesis: the protecting groups

The protein synthesis is highly significant for several reasons like confirming the structure of the natural protein and to investigate the protein structure completely. This function is completely controlled by the amino acid sequence. However, it is not as straight forward like mixing the amino acids together for the formation of the amides. For instances, a mixture of alkaline, A and glycine, G would give the right mixture of amides: A-G, G-A, A-A, and G-G. To control the coupling reaction it is highly important to use some protecting groups.

By protecting the amine groups of one component and the carboxylic groups of the other, particular amide bonds are easily formed. This requires a sequence like,

  • Protect the amino groups in the N-terminal amino acid and the carboxyl group in C-terminal amino acid
  • Couple both the amino acid by the formation of the new amide bond
  • Deprotect the termini of the new peptide and this is required only in some cases

Repeat the process and from that polypeptides can be grown one amino acid residue simultaneously or building pieces and then joining them together.

Application of the custom peptides

Cancer Immunotherapy

This uses the body’s own immune system to attack the cancer cells and to destroy them completely. The peptide-based vaccines use the tumor-associated antigens, which are associated with the T cells that target cancer.

Venom peptides

Bioactive peptides are the highly dominant component of animal venoms. Also, venom peptides are in different lengths and complexity also differs. Thus their synthesis requires a combination of the chemicals and recombinant synthesis to be offered by the GenScript.

Self-assembling peptides

Peptide syntheses are the short synthetic characterized by amphipathic sequences. These peptides will also be able to spontaneously self assemble in aqueous solutions to form highly organized structures like hydrogels.
